The Duolink Solution

The Duolink® kit is based on in situ PLA®, a proximity ligation assay technology, and consists of customisable kit components which can be configured to meet specific assay requirements.

In addition to the kit components you will also need two primary antibodies raised in differents species.

Building a kit that perfectly matches your need is easily done in four steps:

1. Choose PLA probe PLUS against the species one of your primary anibody has been raised in or Probemaker PLUS
2. Choose PLA probe MINUS against the species the other primary antibody has been raised in or Probemaker MINUS
3. Choose detection reagents depending on your mode of detection, fluorescence or brightfield, and filter sets
4. Choose the optimal accessories for your Duolink application

The complete kit contains all buffers and enzymes needed for performing an in situ PLA, except for primary antibodies. You may choose from reagent kit sizes of 30, 100, 500 or 1000 assays of 40µl each. The choice of PLA probes, depends on your primary antibodies and the application you have chosen.

Duolink® Probemaker is a product enabling quick and convenient conjugation of the PLA oligo arms directly to primary antibodies. Directly conjugating primary antibodies enables same species monoclonal or polyclonal use in a Duolink assay. A consequence of this feature is the possibility to study homodimeric protein complexes – an ability unique to Duolink.
Simply include Probemaker probes instead of PLA probes in a Duolink kit.
